What do you mean, "run the file"? Are you referring to playing an MKV file, or starting MakeMKV?

The fact that you said "backup" means you probably used the "backup" function, which creates an image (optionally decrypted) of the Bluray data on the disk.
The next question is, what operating system, and what software do you have installed?
Windows prior to 10 has no support built in for playing BD files. Programs like VLC from Videolan.org do recognize unencrypted Bluray folders. Some media players can handle BD folders, as well. Smart TVs.... BD-aware ones might exist, but I have not seen one myself.
