Yes, but if you "open" the disc like that, the "backup" icon gets greyed-out and you can't make a backup from the backup, which I believe is what the OP was trying to do (for some reason).

If you trick MakeMKV into thinking it's an optical drive by creating a virtual disc, then you can make a backup from a disc image that was already copied.
