I ripped videos to MKV and attempt to play back on DLNA to Panny does not play. "Cannot Play" message comes up.
What could be the problem?

Smart TVs aren't very. DLNA has lots of limitations. That's just two major groups of problems.
One limitation I encountered with DLNA is that it ignored different things according to what server software you used and what client software. Some of my MP4 files would only be seen and played by the client if their file extension was .M4V, while others insisted on .MP4. MKV files were not even shared unless I changed the extension to .AVI, and some clients (Samsung) had no problem with that.
