I do not know how to make a ISO to MKV with DTS option 5.1, please help, thanks.

Step-by-step:
File->Open and navigate to the ISO file.
Select the title(s) and tracks within the titles that you want.
Tell MakeMKV where to save the results ("Output folder")
Set the Properties for the disk to the name of the movie. If you enable Expert mode, you can also set the file name for the movie at this time.
Click the "Make MKV" button.
General steps:
Play with it. You can't break much, as long as you don't overwrite the source ISO. Try selecting different audio tracks. Play with the Profiles (again, you need Expert mode enabled in preferences).

After you open the disk, on the left side you see the list of titles. Next to each one is a triangle... RIGHT click on the triangle, and you will see the tracks that make up each title. Make sure the DTS track is selected.
The normal selection rules for MakeMKV won't select DTS-HD and similar tracks that are "problematic" to play, but you can change that. The rules I use in my normal profile say to select ALL audio and subtitle tracks, because I process the files afterward with handbrake, to make sure I get the "correct" tracks.
