can I use makemkv to create a lossless file for my local server to watch it on my smart TV?

If you purchased the video on optical media, the "lossless quality" ship has already sailed into the sunset... You have a lossy copy of the original video in the package you purchased.

That said, MakeMKV copies the video from the disk to your system without modifying it, so it's as "lossless" as you can get.
