I don't understand the question. If you're asking whether MakeMKV is a 64-bit application, the answer is "now" or "soon" depending on platform. MakeMKV is 32/64-bit on Windows and Linux and 32-bit only on Mac OSX.

That is what I meant: is mkv a 64-bit application? I am a Mac user, is it possible to "recompile" the source codes to make it a 64-bit application? Or, is such a plan to do so for Mac users?

Presently on Mac MakeMKV is a 32-bit Carbon application. The next version (available in a few days) will be still 32-bit, but a Cocoa application. It's certainly possible to produce 64-bit version, however I'd like to know why do you ask? There are compelling reasons to have 64-bit versions on Windows and Linux; but Mac has a different architecture and I don't know any strong argument for 64-bit version. What would be the benefit?

Since using 64bit Handbrake (over 32bit) on my MBP core 2 encoding times (and processor load) have drastically decreased.
Wouldn't that also account for a 64bit makemkv?

No, MakeMKV does no encoding. It's a decryption and copy tool, so something like a faster BR drive or hard drive would make it quicker.
