Galaxy Tab cannot play an MKV file without extra software. There are several free and inexpensive video players available in the Google Play store that can do it.
However, you probably should go over to handbrake.fr and download Handbrake. Start with the release version, but you will eventually want to use the "nightly build" versions, because they have better support for things like Bluray, and better pre-set profiles for Android devices. Android can play MP4 files directly, so long as they meet certain criteria.
