MakeMKV's folder backup option is the 2nd button at the top. Looks like a folder with an arrow. Click it, then check the decrypt box that pops up and choose the location for the backup.

It is best not to play any UHD discs directly. You will wear out your drive's laser fast! Better off ripping the complete disc to a decrypted folder backup and playing that. The Vantec's circuitry has probably died. You could get a cable to run the drive directly, without the enclosure and see if it works or get yourself a new enclosure (I can recommend the icy box enclosure: Icy Box IB-AC640-C3) and see if it works. Since you've had the drive for a few years, it is possible the drive's optics are a little dirty. You could try gently cleaning them with isopropyl alcohol and lint-free cloths.
