Just to add to this, ran into the same problem with Windows 10. Just adding the makemkv.exe file alone didn't fix it, I added the rest of the exe files in the folder as well. Windows Defender Security Center -> Virus & Threat Protection -> Ransomware Protection -> Allow an App through Controlled...
